Description

A solid phase Sandwich ELISA from Assay Genie designed for the quantitative measurement of Selenium-binding protein 1 (SBP1) in mouse samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich detection method and reports a sensitivity of 46.875 pg/mL with an assay range of 78.125-5000 pg/mL. Reagents are intended for storage at 2-8°C for 12 months.

The assay procedure involves equilibration of reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution is added. Reported recovery ranges are 86-104% for serum (average 93%), 85-105% for EDTA plasma (average 97%), and 86-104% for heparin plasma (average 95%). Reported linearity at 1:2, 1:4 and 1:8 dilutions spans 92-104%, 86-104% and 89-96% for serum; 82-97%, 84-95% and 82-96% for EDTA plasma; and 82-90%, 80-93% and 89-98% for heparin plasma.

SBP1, also known by synonyms including SELENBP1, hSBP, hSP56, LPSB, SBP, SBP56 and SP56, has been described in relation to selenium metabolism and is associated with oxidative stress, inflammation and cancer development, making it a candidate biomarker in these areas of mouse research. Sample preparation guidance covers serum (clot and centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant and cell lysate workflows.
