The protocol comprises six steps: reagent and plate preparation with TMB substrate equilibrated to room temperature; primary incubation at 37°C for 90 minutes; detection antibody binding with a biotin-labeled antibody at 37°C for 60 minutes; HRP-Streptavidin binding at 37°C for 30 minutes; color development with TMB substrate in the dark for 10–20 minutes; and stop reaction followed by absorbance reading at 450 nm.
