Validated linearity values are 85-104% in serum, 82-100% in EDTA plasma, and 80-100% in heparin plasma across serial dilutions. Spike-and-recovery averages are 97% in serum, 97% in EDTA plasma, and 96% in heparin plasma. The protocol involves incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution.
