Description

Manufactured by Assay Genie, this Sandwich ELISA kit uses a double-antibody format and colorimetric detection to quantify Arginyl-tRNA Synthetase (RARS) in mouse-derived samples. The assay is reactive with mouse RARS and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Synonyms listed for the target analyte include RARS, ArgRS, DALRD1, Arginine-tRNA ligase, and ArgRS, and the kit is catalogued for metabolism research. Unopened kits are stored at 2–8°C for 12 months.

The assay has a reported sensitivity of 0.469 ng/mL and an analytical range of 0.781–50 ng/mL. In dilution linearity testing (n=5 per matrix), serum recoveries spanned 85–103% at 1:2, 95–105% at 1:4, and 86–104% at 1:8; EDTA plasma spanned 82–96%, 83–99%, and 89–99%; and heparin plasma spanned 81–98%, 83–93%, and 81–100%. Spike-and-recovery testing (n=5) gave average recoveries of 92% in serum (range 86–96%), 97% in EDTA plasma (range 91–104%), and 94% in heparin plasma (range 91–102%).

The workflow comprises reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and stop-solution addition with immediate absorbance reading at 450 nm. Sample preparation guidance includes clotting serum and centrifuging at 1000 × g for 20 minutes; collecting plasma with anticoagulant and centrifuging at 1000 × g for 15 minutes at 2–8°C; homogenising tissue in PBS with protease inhibitors; clarifying cell culture supernatant by centrifugation at 2500 rpm for 5 minutes; and lysing cells with a lysis buffer containing protease inhibitors.
