Mouse RANTES/CCL5 ELISA Kit

From Assay Genie, this Sandwich ELISA kit uses a double-antibody format to quantify RANTES (CCL5), a chemokine involved in inflammation and immune cell recruitment, in mouse samples. It is designed for the precise measurement of RANTES (CCL5) levels and is suited to research into inflammatory and autoimmune conditions and viral infections, where monitoring this chemokine may aid understanding of the immune response and the development of potential therapies.

Reported sensitivity is 9.375 pg/mL with a working range of 15.625–1000 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is reactive with mouse samples and should be stored at 2–8°C for 12 months. Reported linearity for serum (n = 5) spans 86–102% at 1:2, 93–105% at 1:4 and 92–102% at 1:8, while EDTA plasma (n = 5) spans 86–98%, 93–101% and 92–99% across the same dilutions, and heparin plasma (n = 5) spans 86–99%, 85–101% and 82–99%. Reported recovery averages 96% for serum (range 85–102%), 97% for EDTA plasma (85–98%) and 98% for heparin plasma (88–104%).

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks on the pre-coated plate, incubating at 37°C for 90 minutes for antigen binding, adding a biotin-labelled detection antibody with a 60-minute incubation, adding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, then adding stop solution and reading absorbance at 450 nm.
