Mouse Pyruvate Dehydrogenase Alpha/PDHA ELISA Kit

Assay Genie provides a Sandwich ELISA platform for the quantitative measurement of Pyruvate Dehydrogenase Alpha (PDHa) in mouse samples. The immunoassay enables the in vitro quantitative determination of Mouse PDHa concentrations in serum, plasma and other biological fluids, and is offered for research use only.

The assay reports a sensitivity of 0.094ng/ml across a working range of 0.156-10ng/ml. Recovery was evaluated by spiking known levels of Mouse PDHa into serum, EDTA plasma and UFH plasma. Average recoveries were 92% for serum (n=5, range 86-97%), 98% for EDTA plasma (n=5, range 94-105%) and 95% for UFH plasma (n=5, range 86-104%). Linearity was demonstrated by serial dilution of spiked samples, with serum dilutions at 1:2, 1:4 and 1:8 recovering 91-102%, 90-104% and 86-104% respectively, EDTA plasma dilutions recovering 83-101%, 83-96% and 82-98%, and UFH plasma dilutions recovering 87-94%, 84-98% and 87-96%. Intra-assay precision is CV <8% and inter-assay precision is CV <10%.

Pyruvate dehydrogenase E1 component subunit alpha is the mitochondrial somatic-form enzyme (UniProt P35486, gene Pdha1, NCBI Gene ID 18597, molecular weight 43,232 Da) that catalyses the overall conversion of pyruvate to acetyl-CoA and CO2, linking the glycolytic pathway to the tricarboxylic acid cycle. Kit components are stored at 4°C for 6 months.
