Description

A sandwich ELISA from Assay Genie is designed for quantitative detection of mouse PYGM, also known as myophosphorylase or the muscle form of glycogen phosphorylase. The double-antibody assay uses a colorimetric readout measured at 450 nm.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.188 ng/mL with a working range of 0.313–20 ng/mL. The kit is stored at 2–8°C for 12 months. PYGM (UniProt Q9WUB3) is a key enzyme in glycogen metabolism in muscle cells, and its dysregulation has been implicated in muscle disorders such as McArdle disease, making it a relevant biomarker for metabolic research.

Reported assay linearity (n=5) is 95–101%, 87–102% and 99–104% for serum at 1:2, 1:4 and 1:8 dilutions; 82–98%, 84–100% and 82–100% for EDTA plasma; and 84–98%, 91–94% and 81–99% for heparin plasma. Spike-recovery averages are 98% (serum, range 86–99%), 92% (EDTA plasma, range 88–101%) and 97% (heparin plasma, range 92–99%). The workflow includes reagent equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution.
