Mouse PXR/NR1I2 ELISA Kit

A sandwich ELISA, double-antibody assay from Assay Genie designed to quantify the pregnane X receptor (PXR, also known as NR1I2; Uniprot O54915) in mouse samples. The kit is reactive with mouse and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PXR is a nuclear receptor that regulates the expression of drug-metabolizing enzymes and drug transporters, and is involved in the detoxification and elimination of xenobiotics as well as in the regulation of various physiological processes. The kit is intended for research applications such as studying the role of PXR in drug metabolism, xenobiotic response, and disease.

Linearity (n = 5) at 1:2, 1:4, and 1:8 dilutions is 86-101% / 85-97% / 80-99% for serum, 88-103% / 82-99% / 80-99% for EDTA plasma, and 88-100% / 86-101% / 82-99% for heparin plasma. Spike-and-recovery averages 95% across serum, EDTA plasma, and heparin plasma, with recovery ranges of 89-102%, 88-99%, and 86-102% respectively. All reagents are stored at 2-8°C for 12 months.

The colorimetric assay uses a biotin-labeled detection antibody and HRP-Streptavidin (SABC) with TMB substrate, and absorbance is read at 450 nm. Serum is prepared by clotting followed by centrifugation at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C. Cell culture supernatant is clarified at 2500 rpm for 5 minutes, and cell lysates and tissue homogenates are processed with protease inhibitors.
