Mouse Ptpn1 ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of Ptpn1 (Protein Tyrosine Phosphatase Non-Receptor Type 1, also known as PTP-1B and PTP1B) in mouse samples. It uses a double-antibody sandwich format with colorimetric detection, employing a biotin-labeled detection antibody coupled to HRP-Streptavidin (SABC) and TMB substrate, with absorbance read at 450 nm after addition of stop solution.

The assay is reactive with mouse samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.469 ng/mL with a working range of 0.781–50 ng/mL. Linearity in serum (n = 5) is reported as 86–102% at 1:2, 85–103% at 1:4, and 87–102% at 1:8; in EDTA plasma (n = 5) 86–98%, 83–98%, and 86–101% across the same dilutions; and in heparin plasma (n = 5) 82–96%, 82–94%, and 81–95%. Recovery averages are 95% for serum, 96% for EDTA plasma, and 93% for heparin plasma, with corresponding recovery ranges of 94–97%, 91–99%, and 88–98%. The kit should be stored at 2–8°C for 12 months.

The workflow follows a six-step protocol: equilibrate reagents and the TMB substrate to room temperature; run primary incubation at 37°C for 90 minutes; perform detection antibody binding at 37°C for 60 minutes; bind HRP-Streptavidin (SABC) at 37°C for 30 minutes; develop colour with TMB in the dark for 10–20 minutes; and finally stop the reaction and read absorbance at 450 nm.
