Mouse PTN/Pleiotrophin ELISA Kit

Pleiotrophin is a multifunctional growth factor that plays a critical role in cell growth, differentiation, and migration, and is implicated in biological processes such as embryogenesis, tissue repair, and cancer progression. Assay Genie's Mouse PTN/Pleiotrophin ELISA Kit is a sandwich ELISA using colorimetric detection, specifically designed for the precise measurement of pleiotrophin levels in mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The kit offers a sensitivity of 18.75 pg/mL with a working range of 31.25–2000 pg/mL, and components should be stored at 2–8°C for 12 months. Linearity was assessed using serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with serum ranging 89–104%, EDTA plasma 82–101%, and heparin plasma 80–100%. Recovery across samples averaged 97% for serum, 93% for EDTA plasma, and 97% for heparin plasma.

The assay involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labeled detection antibody is then added and incubated for 60 minutes, followed by HRP-Streptavidin for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes before stop solution is added and absorbance is measured at 450 nm.
