Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of intact parathormone (PTH) in mouse samples. Intact parathormone is described as a hormone that regulates calcium and phosphorus levels and plays a role in bone health and mineral metabolism, with dysregulation associated with disorders such as hyperparathyroidism and hypoparathyroidism. The assay is reactive with mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and uses a colorimetric, double-antibody sandwich detection format.

According to the manufacturer, sensitivity is 9.375 pg/mL with a working range of 15.625–1000 pg/mL, and the kit should be stored at 2–8°C for 12 months. Reported linearity values for serum (n=5) span 86–102% at a 1:2 dilution, 85–96% at 1:4, and 80–100% at 1:8; EDTA plasma (n=5) shows 88–105%, 84–101% and 84–92% at the same dilutions, while heparin plasma (n=5) shows 86–102%, 82–100% and 84–100%. Recovery averages 96% for serum (range 88–101%), 99% for EDTA plasma (range 93–105%) and 99% for heparin plasma (range 91–105%).

The procedure involves equilibrating reagents to room temperature, a primary incubation at 37°C for 90 minutes, binding of a biotin-labelled detection antibody at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop step with absorbance read at 450 nm.
