Prosaposin (PSAP) is a 524-amino-acid glycoprotein that is proteolytically processed into saposin A, B, C, and D activator proteins, which stimulate lysosomal hydrolysis of sphingolipids including glucocerebroside, sulfatide, and sphingomyelin. Beyond its lysosomal role, full-length prosaposin is secreted and acts as a neurotrophic factor that supports neuronal survival and myelination, with research interest in neurodegenerative and neuroinflammatory mouse models.

Investigators measuring mouse PSAP typically work with circulating blood fractions or conditioned media from cultured cells, where prosaposin concentrations can reflect altered sphingolipid metabolism or lysosomal storage dysfunction. Common applications include profiling PSAP expression in transgenic mouse models of Gaucher disease, Krabbe disease, and metachromatic leukodystrophy, as well as studying prosaposin signaling in glial and neuronal cell culture systems.
