This quantitative immunoassay from Assay Genie measures endogenous mouse PPP3R1 protein, the regulatory B subunit of calcineurin, in research samples using a 96-assay microplate kit configuration. Calcineurin B (PPP3R1) pairs with the catalytic A subunit (PPP3CA) to form the calcium/calmodulin-activated serine/threonine phosphatase that dephosphorylates NFAT transcription factors, driving cytokine gene expression in activated T lymphocytes and shaping cardiac hypertrophic signaling. Loss-of-function mutations in murine Ppp3r1 have been linked to impaired T-cell development and altered calcium handling in skeletal and cardiac muscle, making reliable protein-level quantitation valuable for immunology, cardiovascular, and neurobiology research programs.

The kit is validated for mouse tissue homogenates and accepts small sample volumes prepared from cultured cells or freshly dissected tissues. Reconstituted reagents should be stored refrigerated and protected from light, with the standard curve generated in the microplate wells provided. The assay uses an absorbance-based readout on a standard plate reader, and unknown samples are interpolated against a dilution series of the supplied reference standard to calculate endogenous PPP3R1 concentrations.

Each kit includes pre-coated microplate wells, biotinylated detection antibody, reference standard, streptavidin-HRP conjugate, and substrate solution sufficient for 96 determinations. The reagents are intended for research use only in laboratory investigations of calcineurin pathway activity.
