Mouse Protein Disulfide Isomerase A3/PDIA3 ELISA Kit

Designed by Assay Genie for the quantification of mouse Protein Disulfide Isomerase A3 (PDIA3), this Sandwich ELISA uses a double-antibody detection format with a colorimetric readout. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and reports a sensitivity of 0.234 ng/mL across a working range of 0.391-25 ng/mL, with reagents stored at 2-8°C for up to 12 months.

Linearity has been demonstrated in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions (ranges of 88-98%, 86-101%, and 81-95% for serum; 93-105%, 83-88%, and 81-100% for EDTA plasma; and 86-105%, 82-101%, and 88-100% for heparin plasma). Recovery ranges of 85-102% (serum, average 93%), 86-105% (EDTA plasma, average 96%), and 86-105% (heparin plasma, average 98%) have been reported. The workflow includes reagent equilibration, sample loading and incubation at 37°C for 90 minutes, biotinylated detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB color development for 10-20 minutes in the dark, and absorbance measurement at 450 nm.

PDIA3, also known as ERp57, is an endoplasmic reticulum chaperone involved in protein folding and quality control, and its dysregulation has been implicated in cancer, neurodegenerative disorders, and autoimmune diseases, making it a relevant biomarker for studying protein folding pathways and disease mechanisms.
