Mouse Prohibitin/PHB1 ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the precise measurement of prohibitin (PHB1) levels in mouse samples including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Prohibitin is a protein that plays a critical role in cellular processes such as cell proliferation, apoptosis, and mitochondrial function, and dysregulation of prohibitin has been linked to cancer, metabolic disorders, and aging-related conditions.

The assay uses a double antibody Sandwich format with colorimetric detection. Sensitivity is 0.234 ng/mL with a working range of 0.391-25 ng/mL. In linearity testing, mouse serum (n=5) gave 89-102% at 1:2, 85-102% at 1:4 and 86-99% at 1:8 dilution; EDTA plasma (n=5) gave 87-100%, 86-101% and 86-96%; and heparin plasma (n=5) gave 89-104%, 85-90% and 84-97% at the same dilutions. Recovery averages were 98% for serum (range 92-104%), 93% for EDTA plasma (range 86-103%) and 95% for heparin plasma (range 85-102%). Kit reagents are stored at 2-8°C for 12 months.

The protocol begins with reagent and TMB substrate equilibration to room temperature, followed by primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB substrate color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after adding stop solution.
