Mouse Progranulin ELISA Kit

Designed for the precise measurement of progranulin levels in mouse serum, plasma, and cell culture supernatants, this sandwich ELISA from Assay Genie uses a double-antibody format with colorimetric detection. It is reactive with mouse samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Progranulin (PGRN) is described as a key protein involved in regulating inflammation, immune response, and cell growth, and dysregulation of progranulin levels has been linked to neurodegenerative disorders, autoimmune diseases, and cancer.

The kit reports a sensitivity of 46.875 pg/mL and a measurement range of 78.125-5000 pg/mL. Linearity across dilutions (1:2, 1:4, 1:8; n=5) is 91-105%, 89-98%, and 82-92% for serum; 90-101%, 84-97%, and 87-100% for EDTA plasma; and 85-104%, 90-97%, and 80-99% for heparin plasma. Spike-and-recovery values are 90-101% (average 95%) for serum, 95-105% (average 100%) for EDTA plasma, and 85-98% (average 91%) for heparin plasma. Components are intended for storage at 2-8°C for 12 months, with several reagents requiring protection from direct light.

The assay procedure includes equilibrating reagents and TMB substrate to room temperature, a 90-minute primary incubation at 37°C, a 60-minute biotin-labeled detection antibody incubation, a 30-minute HRP-Streptavidin (SABC) step, 10-20 minutes of TMB color development in the dark, and absorbance reading at 450 nm after adding stop solution.
