Mouse Pro-NPY/CPON PharmaGenie ELISA Kit (SBRS0073)

A colorimetric Competitive ELISA Kit from Assay Genie, designed for use with mouse samples to measure Pro-NPY/CPON in serum, plasma, and lysate samples. The assay is designed to recognize the C-terminus of pro-NPY and therefore detects both full-length pro-NPY and CPON, including the 69 aa pro-form of NPY. It does not recognize active forms including NPY 1-36, 2-36, 3-36 or 3-35. No cross-reactivity is observed with Ghrelin, Nesfatin, Angiotensin II or APC. The reported detection range is 0.1-1,000 ng/ml with a sensitivity of 0.19 ng/ml or 16.59 pM.

Neuropeptide Y (NPY) is a 36 amino acid peptide hormone involved in obesity, with main effects of increased food intake and decreased physical activity; it also increases the proportion of energy stored as fat and blocks nociceptive signals to the brain. NPY is additionally associated with regulation of energy balance, memory and learning, and epilepsy. Stimulation of NPY activity through certain agonists increases food intake, while blockade of Y1 and Y5 receptors decreases it. NPY synthesis is influenced by glucocorticosteroids and by leptin-related feedback. The 291 bp coding sequence of the NPY gene encodes a 97 aa precursor peptide; cleavage of the N-terminal 29 aa yields pro-NPY (69 aa), which is further processed into NPY 1-39 and the 30-aa C-terminal peptide known as C-flanking peptide of NPY (CPON). NPY 1-39 is further processed to the mature form NPY 1-36. The function of CPON is not currently understood.

For storage and stability, the Pro-NPY Standard, Biotinylated Pro-NPY peptide, and Positive Control should be stored at -20°C after arrival and protected from multiple freeze-thaw cycles; remaining kit components may be stored at 4°C, with opened microplate wells and antibody stable for up to 1 month at 2 to 8°C.
