Assay Genie provides this colorimetric Sandwich ELISA for the quantification of mouse POU domain, class 5, transcription factor 1 (Pou5f1), also known as OCT4. The assay detects natural and recombinant mouse POU5F1 in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, and is intended for research use only in the field of epigenetics.

Reported performance characteristics include a detection range of 0.156–10 ng/mL, a sensitivity of 0.082 ng/mL, an intra-assay CV of 4.2% and an inter-assay CV of 8.4%. Recovery is given as 87% (81–93%) for serum and 89% (83–95%) for plasma. Linearity has been evaluated across dilutions from 1:2 to 1:16 in serum, EDTA plasma and heparin plasma, with reported values generally falling between 82% and 120%.

POU5F1 is a transcription factor that binds the octamer motif (5'-ATTTGCAT-3') and forms a trimeric complex with SOX2 on DNA, controlling the expression of genes such as YES1, FGF4, UTF1 and ZFP206. It is critical for early embryogenesis and for embryonic stem cell pluripotency. Synonyms for the target include NF-A3, Octamer-binding protein 3, Octamer-binding protein 4, Octamer-binding transcription factor 3, Oct-3, Oct-4, OTF-3, Otf-3 and Otf3.
