Description

Manufactured by Assay Genie, this sandwich ELISA kit is intended for the quantitative detection of mouse Potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (HCN4) in serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids, using a colorimetric detection method. HCN4 is a hyperpolarization-activated ion channel with very slow activation and inactivation that exhibits weak selectivity for potassium over sodium ions. It contributes to the native pacemaker currents in the heart (If) that regulate the rhythm of heart beat, may contribute to the native pacemaker currents in neurons (Ih), and may mediate responses to sour stimuli.

Performance characteristics include a detection range of 0.156-10ng/ml and a sensitivity of 0.078ng/mL. Intra-assay precision is 9.1% and inter-assay precision is 11.4%. Reported linearity is provided across dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma, and recovery averages 95% (range 89-101%) in serum and 97% (range 91-103%) in plasma. The assay is specific for natural and recombinant mouse HCN4, also referenced by the synonyms Brain cyclic nucleotide-gated channel 3, BCNG-3 and Bcng3.

HCN4 functions as a homotetrameric complex and is localised to the cell membrane as a multi-pass membrane protein. For research use only; reagent storage conditions differ between components and are detailed in the kit components section. Associated identifiers include NCBI official symbol Hcn4, UniProt accession O70507 and a molecular weight of 127,414 Da.
