Mouse PLA2G2D/Phospholipase A2, Group IID ELISA Kit

From Assay Genie, this sandwich ELISA kit uses a double-antibody method to provide quantitative measurement of mouse Phospholipase A2 Group IID (PLA2G2D) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PLA2G2D is a phospholipase enzyme that catalyzes the hydrolysis of phospholipids to release fatty acids and lysophospholipids and plays a role in the inflammatory response; its dysregulation has been implicated in various inflammatory disorders, making the protein a useful biomarker for research into these conditions and for exploring its potential as a therapeutic target.

The assay shows a sensitivity of 18.75 pg/mL and an assay range of 31.25-2000 pg/mL. Linearity was demonstrated in serum at 87-100%, 85-102% and 91-105% for 1:2, 1:4 and 1:8 dilutions, in EDTA plasma at 82-91%, 82-101% and 85-99%, and in heparin plasma at 86-95%, 82-98% and 86-95%. Spike-and-recovery ranges were 87-104% (average 97%) in serum, 86-98% (average 91%) in EDTA plasma, and 85-97% (average 91%) in heparin plasma. The kit is stored at 2-8°C with a shelf life of 12 months.

After reagent equilibration, standards and samples are incubated on the pre-coated plate for 90 minutes at 37°C, followed by a biotin-labelled detection antibody for 60 minutes at 37°C and HRP-Streptavidin conjugate for 30 minutes at 37°C. TMB substrate is then added and incubated in the dark for 10-20 minutes; the reaction is stopped and absorbance is read at 450 nm. Supplied components include a dismountable pre-coated microplate, lyophilized standard, 100X biotin-labelled antibody, 100X HRP-Streptavidin conjugate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer and plate sealers.
