Description

PICP (Procollagen Type I C-terminal propeptide) is a critical component in the synthesis of collagen, a major structural protein in the body that helps maintain the integrity of connective tissues including skin, bones, and tendons. Abnormal levels of PICP are associated with conditions such as osteoporosis, osteoarthritis, and other collagen-related disorders. The Assay Genie Mouse PICP ELISA Kit is a sandwich ELISA, double antibody assay designed for accurate quantification of PICP in mouse samples, using a colorimetric detection method.

The kit measures levels in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 18.75 pg/mL with a working range of 31.25–2000 pg/mL, and the kit is stored at 2–8°C for 12 months. Linearity across 1:2, 1:4 and 1:8 dilutions is reported as 90–104%, 88–103% and 87–100% for serum (n=5), 82–98%, 83–99% and 88–100% for EDTA plasma (n=5), and 84–94%, 85–98% and 81–92% for heparin plasma (n=5). Recovery averages are 98% for serum (90–103% range), 97% for EDTA plasma (93–101% range) and 90% for heparin plasma (85–96% range).

The assay workflow includes equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution.
