The assay accepts serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, and shows specificity for natural and recombinant mouse PIK3CD.

Performance characteristics include a detection range of 0.312-20ng/mL, a sensitivity of 0.184ng/mL, an intra-assay CV of 6.8% and an inter-assay CV of 8.4%. Average recovery is 89% (range 83-95%) in serum and 91% (range 85-97%) in plasma.

PIK3CD is a phosphoinositide-3-kinase that phosphorylates PtdIns(4,5)P2 to generate phosphatidylinositol 3,4,5-trisphosphate (PIP3), recruiting PH domain-containing proteins such as AKT1 and PDPK1 to the membrane and activating signalling cascades involved in cell growth, survival, proliferation, motility and morphology. The protein forms a heterodimer of a catalytic subunit PIK3CD and a p85 regulatory subunit (PIK3R1, PIK3R2 or PIK3R3), is localised to the cytoplasm, and mediates B-cell receptor and T-cell receptor signalling, NK cell development and neutrophil chemotaxis.
