Mouse Perforin 1 ELISA Kit

A sandwich ELISA kit from Assay Genie designed for the precise measurement of perforin-1 levels in mouse serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Perforin-1 (UniProt P10820; synonyms PRF1) is a key component of the immune system, playing a crucial role in the destruction of target cells by cytotoxic lymphocytes, and its dysregulation has been linked to various immune-related disorders. The kit is suited to research in immune responses, autoimmune diseases, and cancer immunotherapy within the cell biology research area.

Linearity has been demonstrated in serum (92-105%, 87-103%, 98-105% at 1:2, 1:4, 1:8 dilutions; n = 5), EDTA plasma (86-101%, 83-94%, 85-100%), and heparin plasma (80-92%, 93-99%, 83-91%). Recovery ranges are 86-105% in serum (average 96%), 88-103% in EDTA plasma (average 93%), and 86-102% in heparin plasma (average 95%). Reagents are stored at 2-8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes at 37°C, then HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing with TMB substrate in the dark for 10-20 minutes, and measuring absorbance at 450 nm after adding stop solution.
