Mouse Pentosidine ELISA Kit

Assay Genie's Mouse Pentosidine ELISA Kit is a competitive, antigen-coated ELISA designed for the quantitative measurement of pentosidine in mouse samples. Pentosidine is a key biomarker linked to oxidative stress and the aging process, and is implicated in age-related conditions such as diabetes, osteoporosis, and cardiovascular disorders. The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, delivering a sensitivity of 3.75 ng/mL across a working range of 6.25-400 ng/mL.

Validated linearity is reported across 1:2, 1:4 and 1:8 dilutions: serum (n=10) 91-105%, 89-101%, 86-102%; EDTA plasma (n=10) 83-96%, 86-99%, 86-96%; and heparin plasma (n=10) 84-99%, 88-105%, 84-100%. Recovery data show averages of 94% for serum (range 85-103%), 94% for EDTA plasma (range 86-105%) and 100% for heparin plasma (range 90-104%). The sealed kit is stored at 2-8°C and should not be cryopreserved, and absorbance is read at 450 nm after TMB substrate development.

The procedure begins with a 30-minute equilibration of the TMB substrate at room temperature, followed by addition of 50 µL of standard or sample and 50 µL of biotin-labeled antibody per well, incubation at 37°C for 45 minutes, washing, TMB development in the dark at 37°C for 10-20 minutes, and addition of stop solution before reading. Sample preparation covers serum, plasma, cell culture supernatant, cell lysates, and tissue homogenates with specific centrifugation conditions detailed in the supplied protocol.
