Mouse PCSK9/Proprotein Convertase 9 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the accurate measurement of PCSK9 (Proprotein Convertase 9) levels in mouse samples. It uses a double antibody format with colorimetric detection, and is suitable for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PCSK9 is an important protein involved in cholesterol metabolism, influencing LDL receptor degradation and regulation, and its dysregulation has been linked to various cardiovascular diseases, making it a valuable biomarker for research.

The assay has a sensitivity of 9.375 pg/mL and a range of 15.625-1000 pg/mL. Reported linearity for serum (n=5) is 88-98% at 1:2, 83-100% at 1:4, and 90-98% at 1:8. EDTA plasma (n=5) shows 85-101% / 93-105% / 89-98% across the same dilutions, and heparin plasma (n=5) shows 89-99% / 85-91% / 89-98%. Recovery ranges are 86-100% (average 92%) for serum, 88-103% (average 96%) for EDTA plasma, and 88-104% (average 93%) for heparin plasma.

The procedure involves equilibrating reagents and TMB substrate to room temperature and loading standards and samples on the pre-coated plate with incubation at 37°C for 90 minutes, followed by biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB substrate color development in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution. Serum is prepared by allowing blood to clot and centrifuging at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C. The kit should be stored at 2-8°C for 12 months.
