Mouse PARM1 ELISA Kit

Assay Genie provides a sandwich ELISA kit developed for the quantitative detection of PARM1, also known as prostate androgen-regulated mucin-like protein 1 homolog (UniProt Q923D3), in mouse samples. The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and employs a double-antibody sandwich format with a biotin-labeled detection antibody, HRP-streptavidin conjugate (SABC) and TMB substrate, with absorbance read at 450 nm. PARM1 is implicated in androgen-regulated gene expression in prostate tissue and is of interest as a candidate biomarker for prostate cancer and related prostate disorders.

The assay reports a sensitivity of 0.469 ng/mL and a working range of 0.781-50 ng/mL. Validation data supplied by the manufacturer include linearity in mouse serum across 1:2, 1:4 and 1:8 dilutions of 86-101%, 85-97% and 92-101% (n=5), EDTA plasma at 90-99%, 85-101% and 91-97% (n=5), and heparin plasma at 95-105%, 91-99% and 91-97% (n=5). Recovery ranges are 85-105% for serum with a 95% average, 92-98% for EDTA plasma with a 96% average, and 85-100% for heparin plasma with a 92% average. All kit components are stored at 2-8°C for 12 months, with selected components additionally suitable for -20°C storage for 12 months.

The protocol proceeds in six stages: reagent and plate equilibration to room temperature; primary incubation at 37°C for 90 minutes; biotinylated detection antibody binding at 37°C for 60 minutes; HRP-streptavidin incubation at 37°C for 30 minutes; TMB colour development in the dark for 10-20 minutes; and a stop reaction followed by absorbance measurement at 450 nm.
