Description

From Assay Genie, this Sandwich ELISA kit is intended for the quantitative measurement of mouse pancreatic prohormone (PPY) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric, double-antibody sandwich format. Pancreatic prohormone PPY is described as a key regulatory molecule involved in the control of appetite and energy metabolism and is studied as a biomarker in metabolic disorders such as obesity and diabetes.

Reported performance figures include a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. Validation data show recovery in serum of 88–104% with an average of 96% (n=5), in EDTA plasma of 91–101% with an average of 97% (n=5), and in heparin plasma of 85–97% with an average of 90% (n=5). Linearity across 1:2, 1:4 and 1:8 dilutions is given as 89–103%, 83–95% and 83–94% for serum, 92–105%, 83–100% and 82–100% for EDTA plasma, and 88–101%, 83–98% and 80–100% for heparin plasma (n=5).

The protocol equilibrates reagents to room temperature, loads standards and samples onto the pre-coated plate, and incubates at 37 °C for 90 minutes. Biotin-labelled detection antibody is added and incubated at 37 °C for 60 minutes, followed by HRP-Streptavidin (37 °C, 30 minutes) and TMB substrate developed in the dark for 10–20 minutes. The reaction is stopped and absorbance is read at 450 nm. Reagents are stored at 2–8 °C and are stated to remain stable for 12 months under those conditions.
