Description

Assay Genie's Mouse PACAP-38 (Pituitary Adenylate Cyclase Activating Polypeptide-38) ELISA Kit is a sandwich ELISA, double antibody assay designed for the accurate and sensitive detection of PACAP-38 levels in mouse samples, intended for a variety of research applications. The kit offers high specificity and precision, providing reliable and reproducible results.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a sensitivity of 4.688 pg/mL with a working range of 7.813-500 pg/mL, and reagents are stored at 2-8°C for 12 months. Linearity testing across serum, EDTA plasma, and heparin plasma samples produced dilution recoveries of 87-104% to 87-105% (serum), 86-100% to 83-101% (EDTA plasma), and 85-99% to 80-94% (heparin plasma) at 1:2 to 1:8 dilutions. Spike-and-recovery across the same matrices averaged 94% (serum), 100% (EDTA plasma), and 98% (heparin plasma).

The protocol involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, color development with TMB in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution addition. PACAP-38 is a neuropeptide involved in neuronal differentiation, neurotransmitter release, and neuroprotection, with dysregulation implicated in a range of neurological disorders.
