Description

A colorimetric Sandwich ELISA designed for the quantitative detection of oxidized low-density lipoprotein (OxLDL) in mouse samples, manufactured by Assay Genie. The kit is intended for research use and supports studies in cardiovascular disease, particularly atherosclerosis, where OxLDL is a key biomarker associated with plaque buildup in arterial walls.

The assay is configured as a Sandwich ELISA using a double antibody format, with reactivity specific to mouse. It accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.188 ng/mL and the detection range is 0.313–20 ng/mL. Storage is at 2–8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate, incubating at 37°C for 90 minutes, adding a biotin-labeled detection antibody with incubation at 37°C for 60 minutes, adding HRP-Streptavidin (SABC) with incubation at 37°C for 30 minutes, developing colour with TMB substrate for 10–20 minutes in the dark, stopping the reaction and reading absorbance at 450 nm. Sample preparation guidance includes clotting and centrifugation for serum at 1000 × g for 20 minutes, anticoagulant collection and centrifugation for plasma at 1000 × g for 15 minutes at 2–8°C, homogenization in PBS with protease inhibitors for tissue, and clarification at 2500 rpm for 5 minutes for cell culture supernatant. Reported linearity for serum spans approximately 85–105% at 1:2 and 86–101% at 1:8, with EDTA plasma at 82–95% at 1:2 and 82–98% at 1:8, and heparin plasma at 80–100% at 1:2 and 82–100% at 1:8. Reported recovery averages 96% for serum, 93% for EDTA plasma and 96% for heparin plasma.
