Description

From Assay Genie, this Sandwich ELISA (Double Antibody) kit enables quantitative measurement of Cross-linked N-telopeptide of Type I Collagen (NTXI) in mouse samples. NTXI is a key marker of bone resorption that reflects the breakdown of collagen in bone tissue. By measuring NTXI levels, researchers can gain insights into bone turnover and assess interventions aimed at bone-related diseases such as osteoporosis and osteoarthritis, supporting studies in bone metabolism and the development of new therapeutic strategies.

The colorimetric assay has a sensitivity of 1.875 ng/mL and a measuring range of 3.125–200 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2–8°C for 12 months. Recovery and linearity have been characterised in serum, EDTA plasma, and heparin plasma matrices.

The workflow follows a standard sandwich protocol: equilibrate reagents and TMB substrate to room temperature, then load standards, samples and blanks onto the pre-coated plate and incubate at 37°C for 90 minutes to allow antigen binding. Add biotin-labeled detection antibody and incubate at 37°C for 60 minutes, then add HRP-Streptavidin (SABC) and incubate for 30 minutes. Develop with TMB substrate in the dark for 10–20 minutes, add stop solution, and read absorbance at 450 nm immediately.
