Mouse Neurokinin A/NK2R ELISA Kit

From Assay Genie, this sandwich ELISA in a double-antibody format enables quantitative measurement of neurokinin A in mouse samples. The Mouse Neurokinin A/NK2R ELISA Kit is reactive with mouse and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The protocol begins with equilibrating reagents and TMB substrate to room temperature, loading standards, samples and a zero control onto the pre-coated plate, and incubating at 37°C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody is then added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes before the stop solution is introduced and absorbance is read at 450 nm. Recommended sample preparation includes clotting serum (centrifugation at 1000 × g for 20 minutes), collecting plasma with anticoagulant (1000 × g for 15 minutes at 2–8°C), homogenising tissue in PBS with protease inhibitors, centrifuging cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells using a buffer containing protease inhibitors. Linearity at 1:2 dilution is 90–103% (serum), 89–99% (EDTA plasma) and 82–100% (heparin plasma), with mean recovery of 95% (serum), 98% (EDTA plasma) and 94% (heparin plasma).
