SIRT7 is an NAD-dependent protein deacetylase that selectively mediates deacetylation of histone H3 at 'Lys-18' (H3K18Ac), a mark linked to control of gene expression. The enzyme acts as a transcription repressor, supports restoration of ribosomal RNA (rRNA) transcription at the exit from mitosis, and may also deacetylate p53/TP53 to promote cell survival. Dysregulation of SIRT7 has been linked to cancer, cardiovascular disorders, and aging.
