Description

Designed for mouse research, Assay Genie's Sandwich (double-antibody) ELISA Kit quantifies Matrix Metalloproteinase 2 (MMP2), an enzyme involved in extracellular matrix remodelling, cell migration and tissue repair. Dysregulation of MMP2 has been linked to cancer, arthritis and cardiovascular diseases. The kit is intended for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, and uses a colorimetric readout read at 450 nm.

Reported performance from Assay Genie includes a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Linearity (n=5) is 82–103% for serum, 80–102% for EDTA plasma and 80–104% for heparin plasma across 1:2 to 1:8 dilutions. Recovery (n=5) averages 91% for serum, 88% for EDTA plasma and 91% for heparin plasma. Kit components include a pre-coated dismountable microplate, lyophilised standard, biotin-labelled detection antibody (100X), HRP-Streptavidin conjugate (100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer and plate sealer, plus a technical manual, stored at 2–8°C for 12 months.

The procedure starts with equilibrating reagents and the TMB substrate to room temperature before loading standards, samples and a zero control onto the pre-coated plate. Plates are incubated at 37°C for 90 minutes for antigen binding, followed by biotin-labelled detection antibody at 37°C for 60 minutes and HRP-Streptavidin SABC at 37°C for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes before the reaction is stopped and absorbance read at 450 nm. Sample handling notes include clotting serum before centrifugation at 1000 × g for 20 minutes, collecting plasma from anticoagulant tubes after centrifugation at 1000 × g for 15 minutes at 2–8°C, homogenising tissue in PBS with protease inhibitors, and clarifying cell culture supernatant at 2500 rpm for 5 minutes.
