Using a double-antibody format with biotin-labeled detection antibody, HRP-streptavidin, and TMB substrate, the assay is read at 450 nm. Reported sensitivity is 46.9 pg/mL and the standard range is 78-5000 pg/mL. Components are stored at 2-8°C for 12 months.

METAP2 is a key enzyme in protein regulation and cellular processes, with dysregulation linked to cancer, neurodegenerative disorders, and inflammatory conditions, highlighting its value as a therapeutic target and biomarker. Validation data include linearity ranges of 92-100% (serum, 1:2), 85-103% (EDTA plasma, 1:2), and 87-105% (heparin plasma, 1:2) at n=5, with average recovery of 93% for serum, 99% for EDTA plasma, and 90% for heparin plasma. The protocol involves primary incubation at 37°C for 90 minutes, detection antibody binding for 60 minutes, and TMB color development in the dark for 10–20 minutes before stop solution and absorbance reading.
