Mouse Metallothionein-1 (Mt1) ELISA Kit

Manufactured by Assay Genie, this sandwich ELISA uses a double-antibody configuration to quantify mouse metallothionein-1 (MT1). The assay has a sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL. It is validated for mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with colourimetric detection via TMB substrate read at 450 nm.

Reagents and TMB substrate are equilibrated to room temperature; standards, samples and a zero control are loaded onto the pre-coated plate and incubated at 37°C for 90 minutes. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added and incubated in the dark for 10–20 minutes; the reaction is stopped and absorbance read at 450 nm.

Linearity at 1:2, 1:4 and 1:8 dilutions is 87–99%, 88–95% and 82–99% in serum, 92–104%, 82–101% and 80–99% in EDTA plasma, and 88–102%, 83–97% and 86–98% in heparin plasma. Recovery spans 92–103% (mean 98%) in serum, 85–104% (mean 96%) in EDTA plasma, and 91–99% (mean 96%) in heparin plasma. The kit is supplied with a dismountable microplate, lyophilised standard, biotin-labelled antibody concentrate (100×), HRP-Streptavidin conjugate (SABC, 100×), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, wash buffer (25×), stop solution, plate sealer and a technical manual, and is stored at 2–8°C for 12 months.
