The assay workflow includes equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, addition of biotin-labeled detection antibody with a 60-minute incubation, HRP-Streptavidin binding for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding the stop solution.
