Description

Assay Genie provides a Sandwich ELISA kit in a double-antibody format for the quantitative detection of MAP3K5/ASK1 in mouse samples, using a colorimetric detection method. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. MAP3K5/ASK1 is described as a key protein involved in the regulation of cellular stress response and inflammation pathways, with roles in cell growth, differentiation, and apoptosis; abnormalities in its expression have been associated with cancer, autoimmune disorders, and neurodegenerative conditions. The associated Uniprot identifier is O35099 and the product falls within the cell death research area.

Reported sensitivity is 9.375 pg/mL with a working range of 15.625–1000 pg/mL. Linearity for serum, EDTA plasma, and heparin plasma across 1:2, 1:4, and 1:8 dilutions is reported between 84% and 101%, and average recovery across these sample matrices ranges from 92% to 95%. The kit is stored at 2–8°C for 12 months.

Sample preparation involves allowing serum to clot and centrifuging at 1000 × g for 20 minutes; collecting plasma with anticoagulant and centrifuging at 1000 × g for 15 minutes at 2–8°C; centrifuging cell culture supernatant at 2500 rpm for 5 minutes; and homogenising tissue or lysing cells in buffer with protease inhibitors. The six-step assay procedure comprises reagent and TMB equilibration, primary incubation of standards and samples at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes at 37°C, HRP-streptavidin (SABC) binding for 30 minutes at 37°C, TMB colour development in the dark for 10–20 minutes, and stop reaction with absorbance reading at 450 nm.
