Description

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative detection of mouse major centromere autoantigen B (CENPB), also known as centromere protein B or CENP-B. The kit accepts serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids, with reactivity specific to mouse. It recognises both natural and recombinant mouse CENPB and is intended for research use only.

The assay delivers a detection range of 0.156–10 ng/mL with a sensitivity of 0.078 ng/mL. Reproducibility is supported by an intra-assay CV of 6.8% and an inter-assay CV of 9.2%. Dilutional linearity has been validated in serum (109–118%, 94–102%, 97–107% and 109–118% at dilutions of 1:2, 1:4, 1:8 and 1:16), EDTA plasma (88–98%, 94–104%, 83–91% and 94–104%) and heparin plasma (93–105%, 95–104%, 102–112% and 109–118%). Average recovery is 105% in serum (range 99–111%) and 107% in plasma (range 101–113%).

CENPB interacts with centromeric heterochromatin and binds to a specific 17 bp subset of alphoid satellite DNA known as the CENP-B box, helping to organise arrays of centromere satellite DNA into higher-order structures that direct centromere formation and kinetochore assembly in mammalian chromosomes. The protein localises to the nucleus at the chromosome centromere and forms an antiparallel homodimer that interacts with CENPT within centromere complexes containing histones H2A, H2B and H4 together with CENPA, CENPC, CENPT, CENPN, HJURP, SUPT16H, SSRP1 and RSF1.
