Description

A sandwich double-antibody ELISA from Assay Genie for the quantitative detection of low-density lipoprotein (LDL) in mouse samples. The colorimetric assay supports research into cholesterol metabolism and cardiovascular health, including studies of atherosclerosis and potential therapeutic interventions.

The kit is reactive with mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. The cited sensitivity is 4.688 ng/mL with a working range of 7.813-500 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions is 85-103%, 87-95% and 85-105% in serum (n=5); 88-99%, 82-101% and 87-99% in EDTA plasma (n=5); and 80-100%, 80-92% and 81-97% in heparin plasma (n=5). Recovery averages 93% in serum (range 89-96%), 96% in EDTA plasma (range 91-101%) and 95% in heparin plasma (range 86-102%). Unopened kit components are stored at 2-8°C for up to 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks, and incubating the pre-coated plate at 37°C for 90 minutes. A biotin-labeled detection antibody is then added for a 37°C, 60-minute incubation, followed by HRP-streptavidin (SABC) for 30 minutes. Color is developed with TMB substrate in the dark for 10-20 minutes, the reaction is stopped, and absorbance is read at 450 nm.
