Mouse IRE1/ERN1 ELISA Kit

A sandwich ELISA, double-antibody assay from Assay Genie designed for the quantitative detection of mouse IRE1 (ERN1), a key regulator of the unfolded protein response pathway implicated in cellular stress response and cell survival.

The kit uses a colorimetric detection method and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 23.438 pg/mL, with a measuring range of 39.063-2500 pg/mL. Linearity across 1:2, 1:4 and 1:8 dilutions is reported as 90-104%, 92-100% and 80-99% for serum (n=5); 87-95%, 84-101% and 81-100% for EDTA plasma (n=5); and 85-102%, 86-101% and 88-99% for heparin plasma (n=5). Recovery is reported as 85-102% (average 98%) for serum, 88-103% (average 98%) for EDTA plasma, and 93-101% (average 98%) for heparin plasma. Dysregulation of IRE1/ERN1 has been implicated in cancer, diabetes and inflammatory disorders.

The protocol comprises reagent and plate equilibration, a 90-minute primary incubation at 37°C, biotinylated detection antibody incubation for 60 minutes at 37°C, HRP-streptavidin (SABC) binding for 30 minutes at 37°C, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after adding stop solution. Reagents are stored at 2-8°C for 12 months.
