This Assay Genie kit ships with a lyophilized recombinant Activin A reference standard that is reconstituted in sample diluent and used to build a seven-point calibration curve by serial two-fold dilution. The supplied microplate is pre-blocked, and the strips are individually separable so researchers can run partial plates when sample throughput is limited, conserving both reagents and standards.

Standard workflow: load 50 µL of standard or diluted specimen per well, incubate at 37°C for 90 minutes, wash with the supplied buffer, add biotinylated detection antibody working solution for a 60-minute incubation at 37°C, wash again, add streptavidin-HRP conjugate, develop with TMB substrate for 15-20 minutes protected from light, stop the reaction, and read absorbance. The kit has been cited in published studies of folliculogenesis, gonadal steroidogenesis, placental development, hepatic acute-phase signaling, and cutaneous wound healing models, where Activin A concentrations in mouse specimens serve as a quantitative readout of TGF-β family pathway activation. Reconstituted standard should be aliquoted and stored frozen at -20°C to -80°C if not used immediately, and repeated freeze-thaw cycles of the detection antibody should be avoided to preserve binding activity.
