Mouse IL-4 ELISA Kit (MOEB0073)

Designed for the quantitative measurement of mouse Interleukin-4 (IL-4), this sandwich ELISA from Assay Genie reacts specifically with natural and recombinant mouse IL-4 and is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids [S1]. The assay uses a colourimetric sandwich ELISA platform [C0][S3] and absorbance is read at 450 nm on a microplate reader [S1]. The standard protocol involves incubation of standards, samples and controls on the pre-coated plate, sequential addition of detection reagents, washing steps, addition of TMB substrate and reading of optical density once the colour reaction is stopped [S1][S2].

Manufacturer specifications report a detection range of 31.2–2000 pg/mL with a sensitivity of 15.73 pg/mL, an intra-assay CV of 4.3% and an inter-assay CV of 6.9% [S1]. Recovery in serum is 87% (range 81–93%) and in plasma 89% (range 83–95%) [S1]. Linearity has been characterised across serial dilutions (1:2, 1:4, 1:8, 1:16) in serum (N=5), EDTA plasma (N=5) and heparin plasma (N=5), with reported ranges such as 97–105%, 90–100%, 107–119% and 96–108% for serum [S1]. IL-4 is a secreted cytokine that participates in B-cell activation, induces class II MHC expression on resting B-cells, enhances IgE and IgG1 secretion and cell-surface expression, regulates the low-affinity Fc receptor for IgE (CD23) on lymphocytes and monocytes, and positively regulates IL31RA expression in macrophages [S1].
