Mouse IgG ELISA Kit

From Assay Genie, this is a Sandwich ELISA, Double Antibody assay for the quantitative detection of mouse IgG (synonym: Immunoglobulin G1). It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and uses a colorimetric detection method with a sensitivity of 0.938 ng/mL and a working range of 1.563–100 ng/mL.

The supplied reagents include a pre-coated dismountable microplate, lyophilised standard, 100X biotin-labelled detection antibody, 100X HRP-Streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, and plate sealers. The protocol involves equilibrating reagents to room temperature, loading standards and samples and incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody with a 60-minute incubation, adding SABC for a 30-minute incubation, developing colour with TMB substrate in the dark for 10–20 minutes, stopping the reaction, and reading absorbance at 450 nm. All components should be stored at 2–8°C, protected from direct light, with the unopened kit stable for 12 months under these conditions.

Assay performance data: linearity at 1:2, 1:4 and 1:8 dilutions was 85–103%, 85–104% and 91–105% for serum; 83–93%, 82–93% and 82–100% for EDTA plasma; and 81–96%, 89–100% and 80–100% for heparin plasma. Recovery in spiked samples ranged from 89–99% for serum, 85–103% for EDTA plasma and 85–104% for heparin plasma, with averages of 96%, 96% and 93% respectively.
