Description

This colorimetric sandwich ELISA from Assay Genie is designed for quantitative measurement of mouse IGFBP-2 (Insulin-Like Growth Factor Binding Protein 2) in serum, plasma and other biological fluids. The stated detection range is 0.31-20 ng/mL, with a sensitivity of 0.19 ng/mL, a sample volume requirement of 100µL per well and an assay time of 4.5h. The manufacturer reports no significant cross-reactivity or interference between mouse IGFBP-2 and analogues.

The microplate is pre-coated with an antibody specific to mouse IGFBP-2. Standards or samples are combined with this antibody, then a biotinylated detection antibody and an Avidin-HRP conjugate are added. Free components are washed away and substrate is added; wells containing mouse IGFBP-2 appear blue, and adding Stop Solution turns the colour yellow. Optical density is measured at 450 nm ± 2 nm and is proportional to mouse IGFBP-2 concentration, which is calculated by comparing sample optical density with the standard curve. As standard-curve values can vary with assay conditions, the operator should establish a standard curve for each test.

Average recovery was 97% in serum, 101% in EDTA plasma and 90% in cell culture media (n=5 each), and reported intra-assay CV values for three samples were 6.93%, 4.66% and 4.38%. The listed research area is developmental biology. The cited UniProt information states that IGFBP-2 inhibits IGF-mediated growth and developmental rates, that IGF-binding proteins prolong the half-life of IGFs and can inhibit or stimulate the growth-promoting effects of IGFs in cell culture, and that IGFBP-2 binds IGF2 more than IGF1.
