Hyaluronan Synthase 2 (HAS2) anchors at the inner face of the plasma membrane and extrudes high-molecular-weight hyaluronan into the extracellular compartment. A colorimetric, microplate-based ELISA kit from Assay Genie enables quantitative measurement of mouse HAS2 protein in research specimens, including sera, plasma-derived samples, and tissue homogenates prepared from cultured cells or harvested organs. HAS2 is one of three vertebrate hyaluronan synthases (HAS1, HAS2, HAS3) and is the isozyme whose genetic ablation in mice produces embryonic lethality from severe defects in cardiac cushion and yolk-sac vascular development, making its quantification central to developmental and cardiovascular research programs.

Supplied as 96 assays and intended for research use only, the kit accommodates standard ELISA workflows in a microplate format, supporting in vitro protocols in which HAS2 protein levels are compared across treatment arms, developmental time points, or genetically modified mouse strains including transgenics and knockouts. The colorimetric readout is compatible with standard benchtop absorbance plate readers, and the procedure avoids radioisotopes while still allowing quantitative inter-assay comparisons.

Practical applications include tracking HAS2 regulation by cytokines, growth factors, and inflammatory mediators; profiling hyaluronan biosynthesis during wound repair; characterizing tumor-stroma interactions in breast carcinoma and other hyaluronan-rich cancers; and quantifying fibrotic remodeling in lung, liver, and kidney disease models. The breadth of compatible sample matrices makes the kit useful for both in vitro cell-based assays and ex vivo tissue analysis from animal research models.
