Description

Designed for the quantitative detection of Histone H4 in mouse samples, this sandwich ELISA from Assay Genie is suited to epigenetics and chromatin biology research. The assay recognises natural and recombinant mouse Histone H4 (encoded by the Hist1h4a gene) and is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids.

Performance characteristics quoted by the manufacturer include a detection range of 0.156-10 ng/mL, a sensitivity of 0.073 ng/mL, an intra-assay CV of 4.5% and an inter-assay CV of 6.2%. Spike-and-recovery in serum gives an average of 101% (range 95-107%), while plasma recovery averages 103% (range 97-109%). Linearity across 1:2 to 1:16 dilutions is reported as 106-118%, 94-104%, 96-106% and 104-114% for serum; 94-104%, 100-109%, 100-110% and 96-106% for EDTA plasma; and 89-98%, 92-101%, 87-99% and 109-119% for heparin plasma.

Histone H4 (UniProt P62806) is a core component of the nucleosome, the histone octamer that wraps and compacts DNA into chromatin and thereby limits DNA accessibility to the cellular machinery. Through this role, histone H4 contributes to transcription regulation, DNA repair, DNA replication and chromosomal stability, and it is subject to post-translational modifications associated with the histone code. Subcellular localisation is the nucleus. The kit is supplied for research use only.
