Mouse Heparanase/HPSE ELISA Kit

A sandwich double-antibody ELISA from Assay Genie enables quantitative measurement of heparanase (HPSE) in mouse samples, supporting research into the enzyme's role in degrading heparan sulfate within the extracellular matrix. Dysregulation of heparanase has been implicated in cancer, inflammation and cardiovascular disorders, making accurate quantification valuable for studying disease progression and therapeutic intervention. The colorimetric kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples, with storage at 2-8°C for 12 months.

The assay reports a sensitivity of 46.875 pg/mL and a standard range of 78.125-5000 pg/mL. Linearity in serum ranged from 88-99% at 1:2, 84-99% at 1:4 and 80-99% at 1:8; EDTA plasma showed 88-104%, 89-101% and 85-95% at the same dilutions, while heparin plasma showed 86-105%, 83-97% and 82-98%. Spike-and-recovery gave averages of 100% (range 90-105%) for serum, 94% (range 91-102%) for EDTA plasma and 96% (range 86-103%) for heparin plasma.

The protocol comprises a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody incubation, a 30-minute HRP-Streptavidin binding step, TMB substrate colour development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Sample preparation guidance covers serum (clot and centrifuge at 1000 × g for 20 minutes), anticoagulant-collected plasma (centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (2500 rpm for 5 minutes) and cell lysate.
