Mouse Haptoglobin ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of haptoglobin in mouse samples. Haptoglobin is an important protein involved in the acute phase response to inflammation and infection, where it binds free hemoglobin to prevent oxidative damage and promote clearance by macrophages. The assay uses a colorimetric detection method in a double-antibody sandwich format and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance specifications include a sensitivity of 7.5 ng/mL and an assay range of 12.5–800 ng/mL. Storage is at 2–8°C for 12 months. Linearity has been validated across 1:2, 1:4, and 1:8 dilutions in serum (n = 5; ranges 85–96%, 90–100%, 85–98%), EDTA plasma (n = 5; 83–96%, 83–91%, 84–100%), and heparin plasma (n = 5; 88–99%, 82–100%, 81–95%). Recovery averages are 95% for serum (range 85–105%), 97% for EDTA plasma (86–104%), and 93% for heparin plasma (88–96%).

The protocol proceeds in six stages: equilibrating reagents and TMB substrate to room temperature and loading standards and samples for a 90-minute primary incubation at 37°C; biotinylated detection antibody binding for 60 minutes at 37°C; HRP-streptavidin (SABC) binding for 30 minutes at 37°C; TMB color development in the dark for 10–20 minutes; and stop solution addition with absorbance measurement at 450 nm.
