Description

A Mouse H-FABP/FABP3 ELISA kit from Assay Genie, configured as a colorimetric Sandwich ELISA with a double-antibody format. It is designed for the quantitative measurement of Heart-type fatty acid-binding protein (H-FABP / FABP3) in mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The manufacturer specifies a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. The kit is intended for storage at 2–8 °C for 12 months.

H-FABP is a biomarker released into circulation following cardiac injury and is relevant to cardiovascular conditions such as acute coronary syndrome and heart failure. The kit is positioned for research applications including early detection and prognosis of cardiac conditions, cardiovascular studies, and drug development in mouse models. Reported linearity at 1:2 dilution is 93–104% in serum, 82–96% in EDTA plasma and 80–93% in heparin plasma (n = 5 each). Recovery averages are 96% for serum, 96% for EDTA plasma and 95% for heparin plasma (n = 5 each), with respective ranges of 86–104%, 91–102% and 87–104%.

The assay procedure begins with reagent and plate preparation, followed by primary incubation at 37 °C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after the stop solution is added. Sample preparation notes cover serum (centrifuge 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge 1000 × g for 15 minutes at 2–8 °C), tissue homogenate (homogenise in PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate (lysis buffer with protease inhibitors).
