Mouse GPX1/Glutathione Peroxidase 1 ELISA Kit

Assay Genie offers a colorimetric sandwich ELISA, double antibody kit designed for the quantitative detection of mouse glutathione peroxidase 1 (GPX1). GPX1 is a key antioxidant enzyme that protects cells from oxidative damage by reducing hydrogen peroxide and lipid hydroperoxides, and its dysregulation has been linked to cancer, cardiovascular disease, and neurodegenerative disorders. The kit supports research into oxidative stress, antioxidant defence mechanisms, and related disease models in mouse systems.

The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Reported performance includes a sensitivity of 2.813 IU/mL and a detection range of 4.688-300 IU/mL. Linearity in serum (n=5) was 85-102%, 88-103% and 91-103% at 1:2, 1:4 and 1:8 dilutions; EDTA plasma (n=5) gave 86-92%, 82-99% and 84-101%; heparin plasma (n=5) gave 88-100%, 86-98% and 81-94%. Recovery averaged 92% in serum (range 86-99%), 99% in EDTA plasma (range 88-105%) and 92% in heparin plasma (range 88-96%). The kit is stored at 2-8°C for 12 months.

The workflow involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm immediately after adding stop solution.
